New Zealand's Stanaway, 25, will join the GTE Pro title-winning #95 line-up that includes his former Formula Renault 3.5 squadmate Marco Sorensen and Nicki Thiim.
Brazilian driver Serra, 33, will partner Darren Turner and Jonny Adam in the #97 car.
In addition to the Le Mans 24 Hours, Stanaway and Serra will also take part in the two preceding WEC rounds at Silverstone and Spa-Francorchamps.
"I’m hugely excited to have both Daniel and Richie on-board with the team this season," said AMR team principal Paul Howarth.
"Both of them bring with them the speed and consistency that we look for in any driver here at Aston Martin Racing."
Former junior single-seater star Stanaway has been part of Aston Martin's WEC set-up for several years, and was part of GTE Pro class-winning crews in Spa in 2015 and Mexico in 2016.
His 2017 programme includes a part-time Supercars seat, as he will contest the Endurance races in the Australian series alongside Cam Waters at Prodrive.
Serra, a long-time Stock Car Brasil frontrunner, has yet to race in the WEC, but has experience of the Aston Martin V8 Vantage GTE, having done a test and development programme with tyre supplier Dunlop.
The announcement means that none of the six drivers AMR fielded in last year's Bahrain rookie test, including British single-seater stars Alex Lynn and Jordan King, are to join the team's 2017 roster.
